This study explored the protective effects of natural compounds from the medicinal plant Cotula cinerea against biochemical changes caused by the pesticide Chlorpyrifos in rats. A total of 30 male Wistar albino rats were exposed to Chlorpyrifos (9 mg/kg/day) and subsequently treated with Cotula cinerea methanolic extract at doses of 100, 200, and 400 mg/kg/day via oral gavage. After 25 days of treatment, blood samples were collected for serum biochemical analysis, and the liver was preserved for relative weight assessment. The methanol extraction process produced a yield of 5.075%, with a phenolic compounds concentration of 0.142 ±0.03 mg AGE/ml (Gallic Acid Equivalents). Chlorpyrifos exposure led to a decrease in body weight and elevated levels of key liver and kidney markers, including AST (Aspartate Transaminase), ALT (Alanine Transaminase), ALP (Alkaline Phosphatase), LDH (Lactate Dehydrogenase), bilirubin, and cholesterol (p < 0.05 vs. control). In contrast, creatinine and uric acid levels were reduced, indicating nephrotoxicity and hepatotoxicity. However, treatment with Cotula cinerea extract showed improvements in liver and kidney functions and restored body weight. The findings emphasize the significant biological activity of secondary metabolites in Cotula cinerea, which played a key role in mitigating the toxic effects of Chlorpyrifos, both in vitro and in vivo.
